Are some of the TAP-tagged proteins functionally impaired and therefore show an artificial binding pattern?
As far as possible, we tested genetically whether the TAP-tagged proteins perform the function of the wild-type protein and did not find evidence that the TAP interfered with the function of the tagged proteins. We found that strains with TAP-tagged Fus3p, Kss1p and Ste5p form mating projections in the same manner as wild-type strains. In strains with TAP-tagged Tpk1p, we deleted both tpk2 and tpk3 and found that the strain is still viable. Similarly, the TAP-tagged Tpk2p strain in which we deleted both tpk1 and tpk3 is viable. It has previously been shown that at least one functional copy of a tpk gene is required for viability.
