Genetic Reagents
Strains were created for each of the 203 regulators in which a repeated Myc epitope coding sequence was integrated into the endogenous gene encoding the regulator. PCR constructs containing the Myc epitope coding sequence and a selectable marker flanked by regions of homology to either the 5' or 3' end of the targeted gene were transformed into the W303 yeast strain Z1256. Genomic integration and expression of the epitope-tagged protein were confirmed by PCR and Western blotting, respectively.
