Histone Methylation

We examined genomic patterns of nucleosome acetylation using antibodies directed against five different forms of acetylated histone residues, H3K4me, H3K4me2, H3K4me3, H3K36me3 and H3K79me3. Below at left are enrichment profiles for specific methylation events at a specific region and at right are profiles of the average gene for each modification. Trimethylation at H3K4, catalyzed by the histone methyltransferase Set1, is localized to the 5' ends of ORFs and strongly correlates with transcriptional activity (top row). Trimethylation of H3K36, catalyzed by the histone methyltransferase Set2, however, is more broadly distribute throughout the ORF, peaking at the 3' end and more poorly correlates with transcriptional activity. In contrast to both, trimethylated H3K79, catalyzed by the histone methyltransferase Dot1, is fairly evenly distribute throughout the ORF and fails to correlate well with transcriptional activity.
