Global Nucleosome Occupancy

In order to map nucleosome occupancy throughout the yeast genome, we carried out immunoprecipitation with antibodies against core histone H3 or histone H4. Histone occupancy at a region around the CPA1 gene is shown at left. A composite of 5,324 genes is shown in the average gene displayed at right. In general, occupancy appears to be reduced within intergenic regions relative to ORFs.

In agreement with previous gene-specific and genome-wide studies, we find that occupancy by nucleosomes is inversely correlated to transcripional activity (H3 occupancy is shown at left).
